主要研究方向
(1) 高通量免疫組庫測序
免疫組庫測序是通過檢測T/B 細胞受體互補決定區(CDR 區)來全面評估免疫系統的多樣性,深入挖掘免疫組庫與健康或疾病的關系。免疫組測序技術是目前免疫系統研究的最前沿、最熱點技術,廣泛應用在疫苗開發、腫瘤免疫治療、自免疫疾病、藥物研發等等醫學研究的各個領域。
我們課題組擁有世界領先的免疫組庫測序與分析技術,能夠實現了對TCR/BCR的七條鍊無偏差、定量檢測。以此基礎技術上,我們和多個國内頂尖醫學團隊建立了牢固、深入的合作的關系,包括呼吸疾病國家重點實驗室(鐘南山院士、陳淩教授)、湘雅二院(陸前進教授、吳海競教授)、上海質子重離子醫院(蔣國良教授)、江蘇省人民醫院(楊濤教授)等,合作發表高水平研究論文多篇、申請科研項目多項。
(2) 分子診斷技術研究
分子診斷技術是指以DNA和RNA為診斷材料,用分子生物學技術通過檢測特定核酸靶标的存在、缺陷或表達異常,從而對人體狀态和疾病作出診斷的技術,其研究對疾病的預防、預測、診斷、治療和預後具有重要意義。
課題組在國内最早開展基于納米技術的高通量、自動化核酸檢測方法的研究。多年來,我們結合微陣列技術、磁性納米顆粒、熒光雜交技術以及單堿基延伸技術發展了多種高通量自動化核酸檢測方法及其相關技術,并取得了一系列重要的研究成果。我們發展的基于磁性納米顆粒的SNP檢測技術被作為标準方法被編入行業工具書,同時獲得國際同行的高度評價。相關研究先後獲得國家973、自然科學基金及其他省部級重點項目支持。
(3) 全自動一體化POCT系統
基于核酸檢測的全自動一體化POCT系統憑借其靈敏度和準确度高、攜帶方便、操作簡單、檢測時間少、對環境無要求及成本低的優點,已經成為體外診斷行業發展的主流。尤其是在後疫情時代,醫療市場對于此類系統的需求非常迫切。
本人在美國Hudsonalpha生物技術中心和iCubate公司工作器件,作為核心團隊成員發展了新一代“全自動多重分子診斷系統”——“iCubate”系統及其配套試劑盒并且獲得FDA認證。課題組還與上市公司萬孚生物合作完成了該系統的系統升級與國産化,并且取得了三類醫療器械認證。目前課題組開展以一體化檢測器件為載體的小型全自動分子診斷系統研究,重點利用光學、電學、磁學檢測技術對DNA、蛋白質等生物大分子進行檢測。該方面研究受到國家重點研發計劃,自然科學基金和湖南省重點專項等資金的支持。
